CELL METABOLISM

Cells process nutrients and maintain a living state through extremely complex succession of cell metabolic reactions. An array of straightforward cell metabolism assays has been developed at Fluofarma. Tailored for high-throughput screening, with required sensitivity and robustness, our metabolic assays enables to identify and characterize compounds either modulating lipid metabolism or neuroendocrine and endocannabinoid systems.

METABOLISM ASSAYS

Quantification of lipid droplets by live content imaging in 3T3-L1 cells differentiated into adipocytes.

Quantification of lipid droplets by live content imaging in 3T3-L1 cells differentiated into adipocytes.

To identify compounds modulating cell metabolism, we perform a range of cell-based assays in differentiated adipocytes or in polarized primary cultures of hepatocytes:

> Lipolysis assay: multiplexed quantification of triglyceride content and glycerol release, combining extracellular and intracellular measurements by live-content imaging.

> High-throughput cell-based assays to assess neutral lipids and phospholipids accumulation, lipid peroxidation, LDL uptake, NADH/FAD content, P-glycoprotein (P-gp) activity, intracellular glutathione (GSH) content, hepatobiliary function, ...

ENDOCRINOLOGY-RELATED ASSAYS

Neuroendocrine system (mainly HPA axis) plays a fundamental role in response to diverse stimuli, thereby regulating many critical specialized cell functions. However, alterations in signal transduction pathways are involved in the etiopathology of some neurological and psychiatric disorders.

To identify potential positive or negative regulators of neuroendocrine functions, Fluofarma offers to perform a series of cell-based assays in appropriate and predictive cellular contexts.

> Glucocorticoid receptor subcellular translocation

> Prolactin release

> CREB phosphorylation
